چکیده

Human noroviruses (HuNoVs) are one of the leading causes foodborne illnesses globally. The viral genome is most essential information for source tracing and transmission pattern monitoring. However, whole sequencing HuNoVs still challenging due to sequence heterogeneity among different genotypes low titer in samples. To address this need, study, Transposase assisted RNA/DNA hybrid Co-tagmentation (TRACE-seq) method was established next generation library preparation HuNoVs. Our data demonstrated that almost (>7 kb) could be obtained from all 11 clinical samples tested. Twelve including GI.3, GI.4, GI.5, GI.8, GII.2, GII.3, GII.4, GII.6, GII.12, GII.13, GII.14, GII.21 were involved. Compared with traditional metagenomics preparation, optimized TRACE-seq greatly reduced interference host’s bacterial RNAs. In addition, sequences can assembled by using less raw sufficient depth along genome. Therefore, high versatility reliability, promising attainment. It particularly applicable viruses a mixed complicated host background unable cultured vitro, like utilized study.

A Microfluidic DNA Library Preparation Platform for Next-Generation Sequencing

Next-generation sequencing (NGS) is emerging as a powerful tool for elucidating genetic information for a wide range of applications. Unfortunately, the surging popularity of NGS has not yet been accompanied by an improvement in automated techniques for preparing formatted sequencing libraries. To address this challenge, we have developed a prototype microfluidic system for preparing sequencer-...

Tn5 transposase and tagmentation procedures for massively scaled sequencing projects.

Massively parallel DNA sequencing of thousands of samples in a single machine-run is now possible, but the preparation of the individual sequencing libraries is expensive and time-consuming. Tagmentation-based library construction, using the Tn5 transposase, is efficient for generating sequencing libraries but currently relies on undisclosed reagents, which severely limits development of novel ...

Optimization and cost-saving in tagmentation-based mate-pair library preparation and sequencing.

In de novo genome sequencing, mate-pair reads are crucial for scaffolding assembled contigs. However, preparation of mate-pair libraries is not a trivial task, even when using one of the latest approaches, the Nextera Mate Pair Sample Prep Kit from Illumina. To reduce cost and enhance library yield and fidelity when using this kit, we have modified the manufacturer's protocol based on (i) varia...

Miniaturization Technologies for Efficient Single-Cell Library Preparation for Next-Generation Sequencing

As the cost of next-generation sequencing has decreased, library preparation costs have become a more significant proportion of the total cost, especially for high-throughput applications such as single-cell RNA profiling. Here, we have applied novel technologies to scale down reaction volumes for library preparation. Our system consisted of in vitro differentiated human embryonic stem cells re...
